我们的 ADP-Glo™ 分析操作流程
- 配制检测缓冲液(20 mM HEPES、0.01% Triton X-100、2 mM DTT,pH 7.5)。
- 用该缓冲液分别配制 4× ATP 溶液、4× 底物/金属溶液(含各激酶反应所需添加剂)以及 4× 激酶溶液。
- 于黑色聚苯乙烯 384 孔微孔板(Corning #3573)每孔依次加入检测缓冲液 5 µL、4× ATP 溶液 5 µL、4× 底物/金属溶液 5 µL 及 4× 激酶溶液 5 µL,室温孵育 1 小时。
- 每孔加入 20 µL ADP-Glo™ 试剂(Promega),孵育 ≥40 min 以终止激酶反应并降解残余 ATP。
- 随后每孔加入 40 µL Kinase Detection Reagent(Promega),继续孵育 ≥40 min,将 ADP 转化为 ATP。
- 使用 EnVision 2104(Revvity)测定各孔化学发光强度,并将发光值换算为 ADP 浓度,以评估激酶活性。

ATP酶活性评估的重要性
ATPase 是一类仅利用水(H₂O)作为反应物、无需其他分子组分即可催化 ATP 水解生成 ADP 与无机磷酸(Pi)的酶。Carna 在体外实验中发现,某些激酶即便在无肽底物存在的情况下,亦表现出显著的内在 ATPase 活性,自发生成 ADP。ADP-Glo™ 激酶检测系统会将激酶反应产生的 ADP 重新转化为 ATP,并通过荧光素酶反应定量,最终以化学发光信号呈现。虽然发光强度被用作激酶活性的读数,但由内在 ATPase 活性产生的 ADP 并不依赖于底物存在。因此,在解读 ADP-Glo™ 实验数据时,务必将 ATPase 活性纳入考量。
Carna在上表中提供了我们对高纯度激酶的ATP酶活性观察数据。这些数据可用于了解ATP酶活性在ADP-Glo™分析中的潜在影响。



即便在无底物的情况下,随着激酶浓度的升高,RLU 亦呈依赖性增长;而在加入底物后,RLU 虽有进一步升高,但所测得的发光信号主要源于激酶水解 ATP 的 ATPase 活性,而非其特异性磷酸化底物的激酶活性。这表明该激酶的 ATPase 活性显著高于其底物特异性磷酸化活性。
即便采用 ADP-Glo™ 以外的方案测定激酶活性,若所用激酶本身 ATPase 活性较强,也可能导致反应体系中 ATP 降解,进而干扰结果。因此,建议对这类激酶辅以可直接检测底物磷酸化的方法(如迁移率变动分析),以排除 ATP 水解带来的偏差。

当 ATP 浓度接近其 Km 值(如 50 µmol/L)时,ATPase 活性引起的水解可导致 ATP 耗竭,使特异性活性信号趋于平台。在此条件下,随激酶浓度升高,底物磷酸化反而无法被检出。因此,必须将酶浓度调整至不造成信号饱和的范围,方能准确测定激酶活性。
