A pseudokinase is a type of protein that resembles a protein kinase in structure but lacks the ability to catalyze phosphorylation, though it retains a role in cell signaling through non-catalytic mechanisms and plays important roles such as signal transduction, scaffolding for protein complexes, allosteric regulation of active kinases, modulating protein–protein interactions and regulation of localization. Pseudokinases constitute about 10% of the human kinome and are implicated in a wide spectrum of diseases, including cancer, metabolic disorders, cardiovascular diseases, neurodegenerative diseases, autoimmune and inflammatory disorders, developmental abnormalities, expanding the potential as drug discovery targets for novel therapeutic strategies that do not rely on enzymatic activity.
