PhosphoSens™ Kinase Assay Support

Kinase assays utilizing PhosphoSens™ technology (AssayQuant Technologies) enable the acquisition of dynamic kinetic data through real-time and time-course monitoring of enzymatic reactions. The peptide substrate incorporates Sulfonamido-Oxine (Sox), a fluorophore that emits fluorescence upon chelation with Mg2+. When phosphorylated by a kinase, the Sox-containing peptide exhibits increased fluorescence, which can be detected over time. This time-resolved fluorescence signal enhances understanding of reaction mechanisms and supports detailed evaluation of enzyme characteristics and inhibitor effects. With a simple protocol and high throughput capability, this platform is also well-suited for drug screening applications.

PhosphoSens™ Assay Principle & Assay Format

PhosphoSens™ utilizes an improved CSox sensor, in which Sox is introduced into a cysteine residue. When the CSox-conjugated substrate is phosphorylated in the presence of kinase, ATP, and Mg2+, a complex is formed through chelation of Mg2+ with the phosphate group and CSox. Upon excitation, this complex emits fluorescence. .By detecting changes in fluorescence intensity over time, the phosphorylation state of the substrate can be monitored in real time.

Real-time kinetic analysis can be conducted using purified kinase enzymes or various biological samples (such as cell lysates or tissue homogenates), enabling IC50 determination, mechanism of action (MOA/MOI) studies, and structure-activity relationship (SAR) research.
PhosphoSens‑Red(Endpoint Time-Resolved Fluorescence (TRF) Assays)
