PhosphoSens™ キナーゼアッセイサポート
PhosphoSens™(AssayQuant Technologies社)技術を用いたキナーゼアッセイでは、酵素反応をリアルタイムかつ経時的に測定することで動的なキネティックデータの取得が可能です。Mg2+をキレーティングすることで蛍光を発するSulfonamido-Oxine(Sox) を組み込んだペプチド基質はキナーゼによるリン酸化で蛍光を発します。このリン酸化による発光を蛍光強度の増加として経時的に検出したデータは反応メカニズムの理解を深め酵素特性の評価および阻害剤効果の詳細解析に役立ちます。シンプルなプロトコールでスループットが高く薬剤スクリーニングにも最適なプラットフォームです。

PhosphoSens™アッセイ原理&アッセイフォーマット

PhosphoSens™ ではSoxをシステイン残基に導入した改良型CSoxセンサーを使用。CSoxを結合させた基質がキナーゼ、ATP、Mg2+の存在下でリン酸化されるとMg2+がリン酸基とCSoxにキレーティングされることにより複合体が形成され、ここに励起光を照射することで蛍光を発する。この蛍光強度の変化を経時的に検出することで、基質のリン酸化状態の変化をリアルタイムでモニターすることが出来る。
